The National Renewable Energy Laboratory (NREL), located in the foothills of the Rocky Mountains in Golden, Colorado, is the nation's primary laboratory for research, development and deployment of renewable energy and energy efficiency technologies. NREL's Biosciences Center has an immediate opening for a full-time (40 hours per week) Postdoctoral Research Fellow. The initial appointment is for one year, with possible renewal for up to three years.
Job Duties
Conduct heterologous expression, purification, and characterization of cellulase, hemicellulase, accessory, and lignin-modifying enzymes for evaluation of synergistic interactions between enzyme activities on multiple biomass feedstocks. These studies will include cloning of genes of interest into suitable expression vectors for protein production in bacterial, yeast, and filamentous fungal systems. Genes examined will cloned in house from various sources as well as provided by external subcontractors and research partners. Additional proteins will be obtained via purification from either native strains or from commercial enzyme preparations. Purified and characterized enzymes will be used in synergy studies on the hydrolysis of pretreated biomass feedstocks, specifically in the context of high solids. Enzyme improvement will be carried out through both rational design and random evolution methods, with screening development as a critical component of this work.
Required Education and Experience
Must be a recent PhD graduate within the last three years.
Additional Required Knowledge, Skills and Attributes
A recent Ph.D. (less than three years) in microbiology or molecular biology with basic knowledge in microbial physiology, biochemistry, and metabolism. The candidate must be familiar with molecular biology tools and techniques including PCR, plasmid cloning, site-directed mutagenesis, and protein assays. Demonstrated proficiency in protein purification, biophysical protein characterization, and enzyme kinetics is required, specifically for cellulases, hemicellulases, or other biomass-digesting enzymes. Familiarity with biomass hydrolysis under high-solids conditions is also critical, including analysis of monomer and oligomer sugar products by HPLC/LC-MS as well as experience in substrate and product inhibition kinetics for biomass digestion enzymes
Preferred Qualifications
Experience in protein expression in heterologous systems, particularly Saccharomyces, Trichoderma, and Aspergillus. E. coli experience may be useful but not required as few of these enzymes express well in prokaryotic systems. Working knowledge of biomass conversion technologies would also be beneficial.
