We are seeking a highly motivated Postdoctoral Research Assistant to investigate the regulation of inflammation by endocytic pathways in macrophages. We recently found that levels of the anti-inflammatory metalloproteinase inhibitor TIMP-3 are regulated by endocytosis and lysosomal degradation. This project will test our hypothesis that the cell surface levels of TNF-α Converting Enzyme (ADAM17), a key metalloproteinase involved in inflammation, are regulated by this endocytic pathway through its interaction with TIMP-3. The work will be carried out in the group of Dr Linda Troeberg and is based at the Kennedy Institute of Rheumatology, Old Road Campus, University of Oxford.
You will develop expertise in monocyte and macrophage, transfection and quantification of cell surface protein levels, as well as study the effects of LRP-1-resistant TIMP-3 mutants on TACE levels and activity. You will take responsibility for the planning and execution of your own experiments, and develop techniques to support your research.
You will hold a PhD (or be near completion) in biochemistry, cell or molecular biology or related disciplines and have previous laboratory experience in protein biochemistry. Previous experience in monocyte and macrophage biology, enzyme kinetics, metalloproteinases and confocal microscopy would be desirable.
This is a full time, fixed term appointment funded by the Kennedy Trust for Rheumatology Research (KTRR) for 24 months.
