Job Description
SUMMARY:
The focus of the position will be to understand the cellular and molecular mechanisms of immunotherapy in bone tumors using mouse models and in vitro assays. A qualified postdoctoral fellow will have a PhD in cell/tumor biology or immunology with interest in developing a research line on tumor and immune cell function in engineered environments (e.g. the bone microenvironment) and incorporate in vivo imaging of cellular processes including cancer invasion (e.g. in bone), cancer-immune cell interactions, metastasis and response to therapy. The postdoctoral fellow will possess knowledge of immunology and cancer biology, animal expertise, fluorescence microscopy (e.g. epifluorescence, confocal, multiphoton-desired) and be an active member of the research team and collaborate with other lead research investigators and the Primary Investigator in carrying out technical aspects of in vivo and in vitro research projects.
KEY FUNCTIONS:
Requires expertise in mouse handling and disease models in small animals, flow cytometry, and work with cell/tissue culture.
1. Exercises considerable imagination, initiative, and inventiveness in planning and executing the details of experiments, and in evaluating and interpreting the results both independently and with research team members and PI.
Have immunology and cancer biology background and experience in molecular biology and protein biochemistry techniques for cell and tissue samples including immunostainings and microscopy as well as molecular cell analyses. Supports in vitro cell experiments and in vivo animal experiments of tumor cells (e.g. prostate and other) and immune cells. Isolates immune cells from mouse spleen; generates genetically modified cell lines by transfection of fluorescent reporters; maintains cell models by phenotyping (flow cytometry) and FACS sorting; performs immunofluorescence and histology analysis of tissue samples. Based on interest development of the position towards intravital microscopy, tumor microdissection, cell isolation from tumors and molecular profiling is possible.
Knowledge in the following procedures are required:
• Microscopy (confocal and/or epifluorescence; optional experience with multiphoton imaging)
• Animal handling, e.g. breeding, surgery, injection (treatment and tumor implantation) and tissue/organ isolation
• Flow cytometry (FACS) and analysis
• Immunofluorescence staining of tissue samples
• Cell/tissue culture and cryostorage
• Histological procedures (e.g. fixation, slicing, staining, sample storage) and Immunohistochemistry
• Optional: Protein biochemistry (immunoblotting, co-immunoprecipitation, protein purification)
2. Communicate information:
• Must be able to communicate results to and understand instructions from the supervisor and principal investigator.
• Clear and accurate communication in English both face-to-face and over the telephone/skype is required.
• A willingness to cooperate with other staff, students, fellows and visitors is essential.
3. Acquire and develop new techniques:
• Must be able to learn and/or establish novel techniques and protocols for new research lines; provide progress reports in an organized, consistent manner, review current and relevant scientific literature for project(s)
4. Laboratory training and management:
• Supervise and train research assistants, students, fellows and/or visitors
• Assist in lab managing lab records and inventories (recording per computer)
• Assists in ordering of material for the laboratory
• Assist in animal and colony maintenance and records
• Must work with other lab members to ensure the consistent flow of work, necessary materials and reagents
5. Computer literacy:
• Must be able to learn and use software related to data acquisition, image analysis and presentation
