Dr. Huang’s lab focuses on predictive design, synthesis and functionalization of protein-based biomaterials for regenerative medicine and sustainable bio-electronics. Current research projects include: (1) injectable hydrogel and stem cell engineering, (2) stimuli-responsive biomaterial design, and (3) flexible electronics. 研究領(lǐng)域Research: The Leptihn Group offers two Postdoctoral Fellowships; one project focusses on the investigation of membrane proteins controlling cellular signalling pathways employing microfluidics and single molecule methods (fluorescence, electrophysiology) together with the University of Edinburgh and the ZJUI (Zhejiang University-University of Illinois Urbana Champagne). The second project involves viruses of microbes (bacteriophages) with aspects on both, function and structure of phage proteins, as well as phage therapy. Experience in imaging and protein biochemistry or microbiology are desired.

郭博士從事干細胞和癌癥的研究,精于人類和小鼠遺傳學,分子生物學,細胞生物學,生物醫(yī)學,及基因組學的方法。他在造血發(fā)育、白血病干細胞以及腫瘤耐藥調(diào)控方面等有重要建樹。他建立了第一個由PTEN缺失導致T細胞急性白血病的小鼠模型,促成臨床發(fā)現(xiàn)只帶有PTEN突變而沒有NOTCH1突變的嬰幼兒T細胞白血病病人。他首次分離了T細胞急性白血病干細胞,揭示出了多因子突變導致白血病干細胞形成的過程,并發(fā)現(xiàn)了mTOR抑制劑的雙重抗癌機制。近來,郭博士課題組從臨床肺腫瘤中分離出腫瘤干細胞,并發(fā)現(xiàn)NOTCH1信號通路調(diào)控肺腫瘤干細胞的自我更新和鉑類耐藥。
Dr. Wei Guo laboratory uses basic biological approaches to understand biomedical challenges and translates its understanding to clinical practice. There are two major research directions: (1) stress response and ex vivo expansion of hematopoietic stem cells; and (2) regulation and targeted inhibition of cancer resistance.

正在開展的課題:
1)腸干細胞穩(wěn)態(tài)平衡、惡性轉(zhuǎn)化及腸道疾病的早期診療。運用生物信息學分析、小鼠模型和腸道類器官,我們將深入研究大腸腫瘤干細胞的惡性轉(zhuǎn)化和腫瘤轉(zhuǎn)移的分子機制,并開展臨床轉(zhuǎn)化研究。
2)干細胞發(fā)育、衰老和融合的信號調(diào)控。實驗室的最新成果揭示了首個永久性的阻斷體內(nèi)細胞融合的動物模型。在此基礎(chǔ)上,我們將進一步探討細胞融合與衰老的分子機理。

細胞通過感知和轉(zhuǎn)導機械力信號來產(chǎn)生恰當?shù)陌麅?nèi)生物化學應(yīng)答。細胞內(nèi)外的機械力都有助于胞質(zhì)分裂的成功,從而使一個細胞分裂為兩個子細胞。研究表明細胞胞質(zhì)分裂的失敗與基因組不穩(wěn)定性、腫瘤進化和衰老緊密關(guān)聯(lián),而胞質(zhì)分裂的成功則與細胞動力學和細胞外基質(zhì)的相互作用密切相關(guān)。我們的研究目的是探究胞質(zhì)分裂在衰老或其它病理狀況下的對細胞外基質(zhì)不同剛度反應(yīng)的機械力學和動力學機制。為此,我的實驗室聯(lián)合采用化學遺傳學、光遺傳學和高分辨率活細胞顯微成像技術(shù)探究胞質(zhì)分裂失敗產(chǎn)生的結(jié)果和胞質(zhì)分裂的機械生物學機制。
Cells sense and transduce mechanical signals to generate appropriate biochemical and cellular response. Both the cellular and extracellular mechanics contribute to the success of cytokinesis, which is the division of one cell into two. Failure of cytokinesis is implicated in genomic instability, tumor evolution, and aging. The success of cytokinesis is linked to the proper interaction of cytokinetic machinery and the extracellular matrix. We aim to study the mechanics and dynamics of cytokinesis in response to different extracellular matrix stiffness during aging or other pathological conditions. To this end, my lab couple chemical genetics, optogenetics and high resolution live cell microscopy imaging to study the mechanobiology of cytokinesis and the consequences of its failure.

The centrosome is an important signalling and microtubule organizing centre in mammalian cells. Centrosomal defects are a common in high grade cancer and have been shown to trigger tumorigenesis in mice. The impact of centrosomal defects in deregulating the cell cycle in cancer is poorly understood. In addition, cancer genome sequencing efforts worldwide show mutations on centrosomal components are common. Using genetic, cell biology and biochemical assays, we aim to study how the changes in centrosomal signalling lead to the deregulation of the cell cycle in cancer.

Salary is commensurate with qualifications and experience, compliant with policies for postdoctoral recruitment in Zhejiang University. Annual pay is in the region of 300,000 RMB.
